ISOLATION, PARTIAL PURIFICATION, AND APPLICATION OF FUNGAL PROTEASES FROM BEAN CROP SOIL
Abstract
- The present study is based on the characterization of 11 and 16 isolated fungi from the bean crop soil in the Vegetable and Fruit Research Development Centre (Hlegu Township) and Myaungtagar Village, respectively. In a total of 27 isolated fungi, 13 fungi showed a clear zone around the colony to determine the preliminary protease-producing fungi. Still, the two selected fungi (V2 and M16) were found to be the highest protease producers. The two selected fungi (V2 and M16) were found as the genus Penicillium sp. The two selected proteolytic fungi were purified using 50% ammonium sulfate precipitation and then dialysis to obtain a partially purified enzyme. When compared with standard molecular weight markers, the relative molecular weight of Penicillium sp. (16) was found to be approximately ≈ < 40kDa, but Penicillium sp. (2) was estimated to be 70kDa. The partial purification fold was 2.16% for Penicillium sp. (2), which will recover 13.05% by dialysis, but Penicillium sp. (16) had a fold of 2.05% which will recover 23.14% by dialysis. In the Lineweaver-Burk plot, the Km values of Penicillium sp. (2 and 16) were shown to be 1.04 g%. Vmax values were 102.04 mMmin-1 of Penicillium sp. (16) and 100.00 mMmin-1 of Penicillium sp. (2). In the removal of blood stain, Penicillium sp. (2 and 16) exhibited complete clearance of the removal of blood stain compared with the control. In the milk clotting test, two selected, partially purified proteases exhibited strong milk coagulation within about 30 minutes. In the degradation of gelatin using waste X-ray films, it was observed that 70 minutes for Penicillium sp. (16) and 120 minutes for Penicillium sp. (2) were required for complete degradation.
